abstract | Processes to prepare and purify decitabine and processes to prepare a crystalline form of decitabine. Pure decitabine Claim 1: A process for preparing decitabine, characterized in that it comprises a) silylating 5-azacytosine in a nitrile solvent to produce a compound of formula (1) wherein each R is independently an optionally substituted C1-6 alkyl group; b) bonding the compound of formula (1) with between about 0.9 and about 1.2 molar equivalents of a compound of formula (2) per molar equivalent of the compound of formula (1) where R1 represents an alkyl group having between 1 and 6 carbon atoms, and where each R2 is independently an optionally substituted acyl or aryl group, in the presence of between about 1 and about 1.2 molar equivalents of a non-metallic Lewis acid per molar equivalent of the compound of formula (1 ), and with an organic solvent, to provide a compound enriched in the anomer b of formula (3); and c) deprotecting the compound of formula (3) to obtain decitabine. |
