abstract | ABSTRACT OF THE DISCLOSURE The disclosure describes a process for the demonstra-tion and determination of a component of the reaction between a bindable substance which is an antigen, a hapten, or a low molecular substance, and a protein capable of binding the bindable substance specifically. The protein can be an antibody or a specific binding protein. The process involves the steps providing a given quantity of binding partner of the component to be determined in a soluble or insoluble form, reacting the component to be determined with its binding partner, and when the binding partner is in soluble form, adding a given quantity of an antibody against the binding partner, the antibody being in insoluble form. The solid phase is separated from the liquid phase and to the solid phase there is added a coupling product of the component to be determined with an enzyme. The coupling product may also be obtained by binding to an enzyme a substance capable of reacting specifically with the component to be determined or its binding partner. The coupling product is used in quantity at least sufficient to react with all of the binding partner of the component to be determined. Finally, the enzyme activity of the liquid or solid phase of the reaction mixture obtained is determined. Test packs for the performance of this process are also described. This enables to demonstrate or determine substances occurring in extremely low concentra-tions in, for example, serum or urine. |
