abstract The present invention relates to a novel process for preparing a strain of advanced microorganisms for the production of 1,2-propanediol by metabolism of a single carbon source, said method comprising the pressure culture of selection in a suitable culture medium comprising a simple carbon source, an initial bacterial strain comprising a deletion of gènetpiA and a deletion of at least one gene involved in the conversion of methyl glyoxal (propanal) to lactate, in order to to evolve in said initial strain one or more genes involved in the biosynthetic pathway of DHAP to methylglyoxal and then 1,2-propanediol to evolved genes with improved 1,2-propanediol synthase activity, then we select and isolate the strain or strains of advanced microorganisms having improved 1,2-propanediol synthase activity. The invention also concerns the initial microorganisms and the evolved microorganisms thus obtained and a process for preparing 1,2-propanediol and optionally acetone by culturing advanced microorganisms.
