abstract The invention discloses a high affinity fusarium specific single-chain antibody and a preparation method thereof, which includes the following steps: A performing genic mutation to a fusarium graminearum cell wall specific antibody CWP2; B building mutation antibody file bases; C utilizing phage display to select and gather the mutation antibody file bases so as to obtain antibody proteins; D performing PhaseElisa analysis on combining capacity of gathered mutants and fusarium graminearum cell wall proteins; E showing combining strength between the mutants obtained in the ELISA identification step D and a parent antibody CWP2 and the fusarium graminearum cell wall proteins; F showing Westernblot analysis between the high affinity antibody identified by the ELISA and the fusarium graminearum cell wall proteins; and G proving that affinity of the high affinity single-chain antibody and fusarium graminearum cell wall proteins is 15 times of the parent antibody CWP2 when the identified high affinity single-chain antibody and the parent antibody CWP2 perform surface plasma resonance detection. The high affinity single-chain antibody enriches scab resistant sources and establishes the foundation for cultivation of wheat scab high resistant varieties.
