abstract The invention discloses a method for preparing L-dihydroorotate by an enzymatic method. The method includes the steps: taking cyanate, L-aspartic acid and aspartate as raw materials and reacting the raw materials under an alkaline condition to form N-carbamoyl-L-aspartic acid; catalyzing the N-carbamoyl-L-aspartic acid by dihydroorotase or a thallus containing the dihydroorotase serving as a catalyst under a weak acid condition to prepare L-dihydroorotate by means of cyclization; and separating, concentrating and crystallizing obtained dihydroorotate enzymatic conversion solution to obtain the L-dihydroorotate. The N-carbamoyl-L-aspartic acid is catalyzed by the dihydroorotase, a raw material preparation and reaction system is simple, products are easily separated, the shortcoming of difficulty in obtaining raw materials in a chemical synthesis method is avoided, and the dihydroorotase is high in activity and short in reaction time.
