abstract The invention discloses a purification method of portunus trituberculatus reovirus. The purification method is characterized by comprising the following steps in sequence: placing portunus trituberculatus died due to illness to high-salinity TNE 2 (Tris-HCl, NaCl, EDTA) buffer solution which is 8 to 10 times of the volume of the portunus trituberculatus, and homogenizing until the portunus trituberculatus is completely broken; respectively feeding the homogenized tissue suspension into centrifugal tubes; centrifuging and collecting the supernate; homogenizing and centrifuging the precipitate obtained in previous centrifuging again; collecting the supernate; combining the two centrifuged supernate; centrifuging for 20 minutes at 4 DEG C under 8000g; centrifuging the supernate for 1.5 hours at 4 DEG C under 35000g; carefully removing the loosen part on the upper layer of the centrifuged precipitate through the TM2 buffer solution in a flushing manner; suspending the white precipitate at the lower layer into the TM2 buffer solution; completely suspending the dipped white precipitate at the lower layer again; centrifuging for 5 minutes at 4 DEG C under 3500g; extracting the supernate; and repeating the previous steps again so as to obtain the compact pure white part from the lower layer, wherein the pure white part is the purified virus particle. The quick purifying method is efficient, quick, simple and convenient, and low in cost.
