abstract | The invention provides a recombinant microorganism for producing cytidine and a method for producing cytidine by using the recombinant microorganism. Among them, cytidine degradation and utilization genes coding for cytidine deaminase, ribonucleoside hydrolase, cytidine/uridine kinase, and nucleoside transporter were knocked out. Meanwhile, key enzymes in the cytidine biosynthesis pathway are overexpressed, including cytidine triphosphate pyrophosphorylase which degrades cytidine triphosphate to cytidine monophosphate, and cytidine monophosphate phosphorylase which catalyzes cytidine monophosphate to cytidine. In addition, the pyrimidine nucleoside pathway is subjected to genetic engineering modification, and the feedback inhibition of the synthetic pathway is relieved. The recombinant strain can reach the cytidine yield of more than 20g/L in a 5-liter fermentation tank by a biological fermentation method, so that the industrial mass production can be realized, and meanwhile, the cytidine has low production cost, less pollution reduction, environmental protection and higher popularization and application values. |
