A preparation method and application of an isourea hapten and an antigen, wherein the isourea hapten is reacted with 3,5-dichlorophenyl isocyanate and glycine ethyl ester hydrochloride to form 2‐(3‐(1,5‐two Ethyl chlorophenylurea) ethyl acetate, hydrolyzed to give 2‐(3‐(1,5-dichlorophenylureido))acetic acid, and then ring-forming to give 3‐(3,5-dichlorobenzene )2,4-imidazolidinyldione, and then reacted with 6-aminohexanoic acid methyl ester obtained by the reaction of methyl 6-aminohexanoate with triphosgene to form 6‐(3‐(3) ,5-dichlorophenyl)- 2,4-dioxyimidazolidinyl-l-carboxamido-hexanoic acid methyl ester, finally obtained by hydrolysis under acidic conditions; iprodione antigen from iprodione hapten and carrier Protein coupling is obtained. The antigen prepared by the present invention exhibits a specific isoficidal antigenic determinant, making it possible to screen for a highly specific isocyanuric monoclonal antibody. The produced antibody has high specificity and high sensitivity, and can be used for rapid determination of enzyme-linked immunoassay and test paper. |
