abstract | The present invention relates to a method for preparing a library of saposin lipoprotein particles, wherein the particles comprise membrane components from cell membranes or organelle membranes and lipid-binding polypeptides belonging to the lipid-interacting protein SAPLIP A family of saposin-like proteins or derivative forms thereof, wherein the method comprises the steps of: a) providing a mixture of crude membrane vesicles derived from cell membranes or organelle membranes; b) subjecting step a) in a liquid environment contacting the mixture with a lipid-binding polypeptide; and c) performing self-assembly of the particle. The present invention also provides a method of preparing purified saposin lipoprotein particles comprising the step of preparing a library according to the above method and the additional step of f) purifying saposin lipoprotein particles from the library. Furthermore, the present invention provides saposin lipoprotein particle libraries and saposin lipoprotein particles obtainable according to the method of the invention. These substances can be used in medicine, in particular for preventing, treating or reducing the severity of disease, or in diagnostic methods, cosmetic treatments or as vaccination preparations or as tools in drug development, drug screening, drug discovery , antibody development, therapeutic biologics development, membrane or membrane protein purification, membrane protein expression, membrane and/or membrane protein research, especially lipidomics and proteomics, preferably membrane and/or membrane protein isolation, identification and/or research or create lipidome or proteome databases. |
