abstract |
The invention relates to and discloses a method for constructing recombinant Escherichia coli with high yield of p-aminophenylalanine. The present invention screened the PAPA biosynthesis pathway from different strains, and determined the pathway source of high-yielding PAPA through combinatorial optimization. The combined pathway was transferred into Escherichia coli, and the host strain was genetically engineered to strengthen the chorismate synthesis pathway and weaken its metabolic pathway. Recombinant Escherichia coli that can finally produce high PAPA is obtained. The PAPA yield of this strain can reach 752.63mg/L. The invention lays a foundation for the industrialized production and application of PAPA. |
