abstract The invention discloses a method for separating and extracting UMP sodium salt, comprising the following steps: (1) adding a phosphatase solution and a catalyst to a catalytic solution containing UMP sodium salt to carry out esterification reaction to obtain an enzyme catalytic solution containing UMP ester; (2) adding the obtained UMP ester-containing enzyme catalytic solution into ethyl acetate for extraction to obtain an ethyl acetate solution containing UMP ester; vacuum concentration to remove the ethyl acetate to obtain a UMP ester concentrated solution; after adjusting the pH, carry out deesterification reaction (3) after adjusting the pH of the obtained reaction solution, it is filtered through a nanofiltration membrane to obtain a nanofiltration retentate; (4) after the obtained nanofiltration retentate is concentrated by evaporation, an anti-solvent is added for crystallization, centrifugation, and drying to obtain UMP sodium salt. The invention adopts the method of extraction after esterification, omits the process of chromatography and solid-liquid separation, and the molar yield of UMP sodium salt is over 92%, and the purity can reach over 98%.
