abstract | The present invention provides a method for screening polypeptides specifically bound by an antibody based on a polypeptide chip and the application of the screened polypeptides. The screening method includes using a peptide chip to screen antibody-containing sample dilutions and antibody-free sample dilutions for the first time under blocking and non-blocking conditions, respectively, to obtain the first seed peptide library under blocking conditions and non-blocking conditions. The first seed peptide library under the two conditions; the second seed peptide library is obtained by taking the intersection of the polypeptides in the first seed peptide library under the two conditions; the serum samples containing antibodies and the antibodies without antibodies were analyzed using a peptide chip under blocking conditions. The serum sample is screened for the second time to obtain the third seed peptide library; the intersection of the third seed peptide library and the second seed peptide library is taken to obtain the fourth seed peptide library; the antibody is one or more single antibodies, or multiple mixed antibodies . The method excludes interfering peptides under different conditions, and the obtained peptide collection has higher specificity, which is suitable for the detection of antibodies in complex clinical samples. |
