abstract |
The invention relates to the synthesis of nucleosides with a fluorescent tricyclic base, their incorporation into nucleic acids and their use in the analysis of said nucleic acids and for stabilizing said nucleic acids against thermal denaturation. The fluorescent nucleosides described in the present invention preferably comprise ethene derivatives of 2'-deoxy-7-deazaadenosine and of 2 ', 3'-dideoxy-7-deazaadenosine and their incorporation into nucleic acids in a chemical or enzymatic manner. The presented invention is particularly useful in the determination of nucleic acids, especially in the field of medicine. The advantage of the presented invention in comparison to others is that now chemically stable, strongly fluorescent nucleic acid building blocks with only little structural modification of natural nucleosides are used for the synthesis of nucleic acid-binding compounds. |
