abstract | The invention provides descriptions of methods and intermediates for the stereoselective alkylation of carbonyl groups. The invention, in particular, allows stereoselectively synthesizing the antidepressant drug escitalopram. According to the invention, it was found that derivatives of boric or boronic acid are suitable bridging elements for attaching a chiral group to a compound containing an alkylated carbonyl group. Thus, these borates and boronates are useful in a method for asymmetrically alkylating a carbonyl group in a compound containing a carbonyl group and a linking group capable of reacting with a boric or boronic acid derivative. Asymmetric alkylation can be carried out by mixing a compound containing an alkylated carbonyl group and a linking group capable of reacting with a boric or boronic acid derivative, adding chiral alcohol and adding an organometallic compound. After the alkylation reaction, borate and boronate can be easily removed by hydrolysis. |
