abstract | The invention relates to a protein or a complex of proteins which stimulate the transcription of activated DNA in eukaryotic cells, to genes encoding the protein or proteins, and to uses thereof. The protein or protein complex, referred to as a transcription adapter, is associated with two other factors, each of which, in turn, binds to a region of a eukaryotic promoter. The first of these factors, called activator, binds to a region of the DNA sequence of the promoter called the UAS binding site or transcription activator. The second of these factors, referred to as general transcription factor (GTF), binds to a second region of the promoter DNA sequence, called the TATA box, located downstream of the UAS binding site. . The transcription adapter specifically binds to an acidic activation domain (i.e. a domain extremely enriched in acidic amino acids, such as aspartic acid and glutamic acid) located on the protein of the activator which further comprises a DNA binding domain. |
