abstract | Methods and constructs are provided for the introduction of multiple genes into plants using a single transformation event. Coordinated expression of genes in the cassette, producing proteins with native amino acid sequences, is achieved by production of one polycistronic mRNA that contains separate translation initiation signals for each enzyme coding region. Bicistronic constructs contain a single 5' promoter, protein encoding sequence 1, an IRES, protein encoding sequence 2, and a single 3' polyadenylation sequence. For polycistronic constructs, additional cassettes of protein encoding sequences, in which each coding region is preceded by an IRES, can be inserted between protein encoding sequence 2 and the polyadenylation sequence. The methods and constructs are useful for creating plants with stacked input traits (e.g., glyphosate tolerant plants producing BT toxin) and/or value added products (e.g., the production of PHAs in plants). |
