This invention relates to a method of analyzing a biological sample, said sample comprising one or more (poly)peptides, said method comprising (i) bringing said biological sample into contact with a cross-linking fixative; (ii) neutralizing any excess of said cross-linking fixative; (iii) adsorbing the sample fixated in (i) on the surface of a solid support; (iv) adding one or more endoproteases; and (v) detecting of one or more cleavage products of said endoprotease(s) by mass spectrometry. Also provided is a method of processing a biological sample, said sample comprising one or more biological macromolecules, said method comprising exposing the sample to elevated hydrostatic pressure and, either concomitantly and/or subsequently, to a cross-linking fixative. Furthermore provided are kits useful for performing the methods of the invention. |
