abstract |
A human anti-IL-12 monoclonal antibody, comprising a p35 subunit and a p40 subunit that form a p75 heterodimer, wherein said monoclonal antibody recognizes the p40 subunit of human IL-12 and neutralizes at least 90% of the bioactivity of Human IL-12 measured by: (a) the inhibition of the proliferation of human lymphoblasts activated by phytohemagglutinin (PHA) stimulated by IL12, in which the concentration of said monoclonal antibody is 0.5 μg / ml and the concentration of said IL -12 human is 0.25 ng / ml or (b) inhibition of IFN-γ production by human lymphoblasts activated by PHA stimulated by IL-12, in which the concentration of said antibody is 0.5 μg / ml and the concentration of said human IL-12 is 0.25 ng / ml and wherein said monoclonal antibody can be obtained from a mouse deficient in a p35 or p40 subunit gene of IL-1-12. |
