abstract | Method for the selection of bispecific single chain antibodies comprising a first binding domain that can bind to a CD3 epitope and a second binding domain that can bind to the extracellular domain of prostate specific membrane antigen (PSMA), comprising the steps of: (a) providing at least three types of host cells that express (i) the human extracellular domain wt of PSMA having SEQ ID: 447 on the cell surface; (ii) a mutated form of human PSMA wt on the cell surface, in which the amino acid residues at positions 140, 169, 191, 308, 334, 339, 344, 624, 626, 716, 717 and 721 are mutated to the corresponding amino acid residues of rodent PSMA wt; and (iii) the wt rodent extracellular domain of PSMA on the cell surface; (b) contacting each type of host cell (i), (ii) and (iii) of step (a) with bispecific single chain antibodies and effector T cells; and (c) identify and isolate bispecific single chain antibodies that mediate lysis of host cells that express the human extracellular domain wt of PSMA on the cell surface according to (b) (i) and host cells that express the mutated form of human PSMA wt on the cell surface according to (b) (ii) but not of host cells expressing the wt extracellular domain of rodent PSMA on the cell surface according to b (iii). |
