abstract | A method of preparing a perfusion-based 3D in vitro cell culture system, comprising: selecting a matrix from a perfusion-decellularized liver or pancreas that has an intact vascular network and a population of stem cells that have the ability to differentiate in a cell type present in a native liver or pancreas; and perfusing the population of cells into the perfusion-decellularized matrix under conditions that allow seeding of the perfusion-decellularized matrix and differentiation and functional maturation of the stem cells in the population to cell types present in the liver or pancreas prior to infusion. decellularization, wherein the conditions also include perfusing the matrix with medium containing differentiation pathway activators or inhibitors selected to provide specific differentiation of liver cells or pancreatic cells. |
