abstract | A method of screening a polypeptide for a desired activity against a target molecule, and the method comprises: a) culturing a Gram negative bacterial cell comprising a polynucleotide encoding the polypeptide so that the polypeptide is produced, b) permeabilizing the membranes Cells of the bacterial cell with a non-ionic detergent or an organic solvent, in which case the polypeptide and the polynucleotide encoding the polypeptide are retained within the permeabilized bacterial cell, c) contacting the permeabilized bacterial cell with the target molecule of such that it diffuses into the permeabilized bacterial cell, and d) screening the polypeptide for the desired activity, in which the polypeptide has a molecular size sufficient to retain the polypeptide within the wall of the bacterial cell, or the polypeptide is associated with at least a second polypeptide to form a protein complex that is retained d within the permeabilized bacterial cell, and the second polypeptide is selected from: i) a polypeptide having a molecular size such that the protein complex is retained within the wall of the permeabilized bacterial cell; and / or ii) a DNA binding protein. |
