abstract |
A cell-free method for producing closed linear deoxyribose nucleic acid (DNA) molecules comprising: (a) contacting a template comprising a linear double-stranded DNA molecule covalently closed at each end by a portion of a sequence of protelomerase recognition and comprising at least one stem-loop motif with a strand displacement polymerase under conditions that promote amplification of said template in the presence of at least one primer that is capable of specifically binding to a primer binding site within said loop stem motif; (b) contacting the DNA produced in (a) with at least one protelomerase under conditions that promote the production of closed linear DNA. |
