abstract | 1325912 Streptokinase - carbohydrate polymer compositions BAXTER LABORATORIES Ino 11 Nov 1970 [2 Dec 1969] 53566/70 Heading C3H A streptokinase composition comprises one part by weight of streptokinase chemically bonded to from one to five hundred parts by weight of a support of a polymer comprising carbohydrate units for example polysaccharides such as cellulose, dextran, starch or dextrins, preferably having a molecular weight of 70,000 to 500,000, polymerized derivatives of carbohydrates, alkali metal containing derivatives of carbohydrates, copolymers of sucrose and epichlorohydrin, carbohydrates etherified with aminoalkyl groups such as aminoethyl and carboxyl containing derivatives for example carboxyethylcellulose, carboxymethyldextran and carboxypropyldextran. In the preferred compositions the streptokinase is bonded to the support via the groups wherein R is the support medium having a bond on an oxygen atom connected to the moiety, R<SP>1</SP> is streptokinase, n is a positive integer and X is a monovalent hydrocarbon radical having no more than 4 carbon atoms, a hydrogen or halogen atom or a further R or R<SP>1</SP> group. The compositions are obtained by reacting the streptokinase with carbohydrate azides, triazine derivatives or by the use of carbodiimides. In the examples "dextran azide" is prepared by diazotization of the hydrazide obtained by reaction of hydrazine and carbomethoxy dextran, or with carboxyethyl dextran. A copolymer of sucrose and epichlorohydrin is converted into a streptokinase reactive compound with cyanuric chloride or a substance of the formula in which X is Br, Me, H, Pri, n-Bu, I, or allyl. |
