The present invention provides families of compounds that can be used as labels (referred to as eTag reporters), linked to polymeric compounds, and then assayed for the release, sequestration, and detection of eTagporters from the polymeric compound. For oligonucleotides, eTagrporters are synthesized at the end of the oligonucleotides using phosphite or phosphate chemistry, and mass modifying regions, charge modifying regions, and detectable regions are sequentially added to different molecules, and eTag-labeled reporter molecules. An eTag reporter can be generated linked to the soligonucleotide of interest for identification. Protocols are used that release the eTag reporter when the target sequence is present in the sample.
