abstract The present invention determines the base sequence of a nucleic acid molecule that utilizes dNTPs that have been labeled at the 3 'end with a cleavable tag that inhibits further elongation and uniquely identifies the base to which they are attached. In a related embodiment, an oligonucleotide having a cleavable tag associated with a base sequence is used in a ligation reaction to determine the base sequence of a particular DNA sample.
