abstract PROBLEM TO BE SOLVED: To provide a novel selection marker gene that can be used for introduction of a heterologous gene using Neisseria gonorrhoeae, a vector containing this marker gene, an Neisseria gonorrhoeae mutant strain that can be transformed using this vector, a method for transforming this mutant strain, Provided is a transformant obtained, and a method for producing a protein using the transformant. A gonococcal leucine-requiring mutant leu-5 (FERM P-20079) and a gonococcal leucine-requiring / nitrite-requiring mutant leuN-5 having high spore-forming ability and good growth were obtained. In addition, leu2 and leu2B were obtained as genes that complement the leucine requirement of the Neisseria gonorrhoeae mutant. Since the mutant strain is sensitive to pyrithiamine, it can be transformed with leu2 or leu2B and ptrA as selection markers, or further with niaD as a selection marker. [Selection figure] None
