abstract | PROBLEM TO BE SOLVED: To provide an efficient mass synthesis method and purification method of CMP-deaminoneuraminic acid (CMP-KDN). In a method for enzymatically producing CMP-deaminoneuraminic acid (CMP-KDN) from cytidine 5'-triphosphate (5'-CTP) and deaminoneuraminic acid (KDN), CMP is used as the enzyme. Provided is a process for producing CMP-KDN using N-acetylneuraminic acid synthase (CMP-NeuAc synthetase). Moreover, the refinement | purification method of CMP-KDN which consists of process 1-4 is provided. (Step 1) A step of adding a divalent cation to a CMP-KDN-containing solution to precipitate coexisting phosphate, pyrophosphate, and nucleotide, (Step 2) a step of converting a nucleotide into a nucleoside by an alkaline phosphatase reaction, (Step 3) ) A step of precipitating CMP-KDN with an organic solvent; and (step 4) a step of recovering CMP-KDN.
