A genetically engineered strain of the oleaginous yeast Yarrowia lipolytica capable of producing over 25% eicosapentaenoic acid (EPA, ω-3 polyunsaturated fatty acid) in the total oil fraction is described. These strains comprise various chimeric genes that express heterologous desaturases, elongases, and acyltransferases, and optionally comprise various natural desaturases and acyltransferase knockouts, enabling EPA synthesis and high accumulation To.
