A) providing a sample containing an RNA molecule; b) hybridizing a first polynucleotide to the RNA molecule; c) reverse-trancribing the RNA molecule to generate first strand cDNA; d ) Hybridizing the second polynucleotide to the first strand cDNA, the second polynucleotide comprising: (i) a 3′-part complementary to a portion of the first strand cDNA (ii) second primer binding A 3′-nonextendible oligonucleotide comprising a 5′-overhang comprising the sequence of the site, e) obtaining an extension reaction product comprising a sequence complementary to the second primer binding site from the first strand cDNA, f A) amplifying the extension reaction product by polymerase chain reaction using a first primer and a second primer complementary to the second primer binding site; g) detecting the amplification product by real-time fluorescence readout; Of RNA molecules, including Way out.
