One aspect of the invention relates to a double-stranded nucleic acid that is useful as a siRNA and has a sense strand and an antisense strand for a target nucleic acid, wherein the sense strand is one with one or more modified nucleobases, or an antisense strand. Contains one or more mismatched base pairings. Another aspect of the invention relates to a single stranded oligonucleotide comprising at least one nucleoside comprising an unnatural nucleobase. Another aspect of the present invention is a method of gene silencing comprising administering to a mammal in need thereof a therapeutically effective amount of a double stranded oligonucleotide containing a sense strand and an antisense strand, The sense strand relates to a method that contains one or more modified nucleobases or one or more mismatched base pairings with the antisense strand. |
