abstract | Methods are provided for qualitatively and / or quantitatively detecting the presence of mutations, modifications or impurities in a protein sample. The method uses an isotope label of an amino acid incorporated into a protein before protein digestion so that the two protein samples can be compared by bottom-up liquid chromatography. The measurement realized by the present invention considers not only the mass of the peptide produced during proteolytic digestion but also the abundance of each obtained peptide. Peptide abundance can be measured by MS using a SITRS sample that is mixed with unlabeled protein prior to protein digestion. As used herein, the term “SITRS sample” or “SITRS standard” means a protein of a known sequence (eg, an antibody) labeled with a stable isotope label of at least one amino acid present in the protein. |
