Novel compositions, uses and methods of making the same are provided. In general, the present invention provides novel quinolone compounds and pharmaceutical compositions thereof that can inhibit cell proliferation and / or induce cell apoptosis. The invention also provides methods for preparing such compounds and compositions, and methods for making and using the same. In certain embodiments, the compound inhibits ribosome biogenesis by inhibiting POL1 transcription, and the disease or condition is susceptible to treatment or prevention by inhibiting POL1 transcription.
