abstract The present invention relates to a solid phase chelating material, particularly a solid phase chelating material useful for purifying proteins. The solid phase chelating material contains a solid phase, a polyamine group bonded to the solid phase, and a chelating group bonded to the polyamine group. At least a part of the polyamine group is bonded to at least two chelating groups for each polyamine group. Each chelating group contains one or several aminopolycarboxylic acid groups (APA groups), and the number of APA groups per polyamine group bonded to at least two chelating groups is at least 3. Some APA may form a linear chelate chain, or the APA group may form a branched chelate chain to which the APA group is interconnected by a bifunctional or trifunctional linker. The solid phase chelating material carrying a metal ion according to the present invention exhibits strong metal bonding, high stability to chelating agents, reducing agents and alkaline solutions, high affinity to proteins, and extremely high binding ability. Furthermore, the present invention purifies a recombinant polypeptide and protein with a histidine tag, a phosphoprotein and a metal coordination protein using the method for producing the solid phase chelating material and the solid phase chelating material carrying a metal ion.
