abstract | (57) [Summary] [Object] An object of the present invention is to provide a method for detecting a polynucleotide suitable for a highly sensitive, highly accurate, quantitative, and high throughput automated apparatus. [Constitution] A fluorescent labeled probe 2 which hybridizes to a target polynucleotide chain 1 is used as a fluorescent labeled probe, and both are reacted to obtain a fluorescent labeled probe 3 bound to the target polynucleotide 1. After unreacted excess fluorescently labeled probe was separated and removed by electrophoresis, the temperature was raised to above the denaturation temperature of the conjugate 3 of the target polynucleotide and the fluorescently labeled probe, and then electrophoresis was continued to bind to the target polynucleotide. The fluorescently labeled probe 4 is released, and the released fluorescently labeled probe 4 is irradiated with the laser light beam 6, and the emitted fluorescence is detected by the fluorescence detector 7. [Effect] There is an advantage that a series of steps can be sequentially and automatically performed. |
