abstract | (57) [Summary]nThe present invention provides a method of preparing a cell population enriched for erythroid progenitor cells (BFU-E) from hematopoietic cells in which up to 100% of the colony forming units are BFU-E. The method involves the separation of hematopoietic progenitor cells based on binding to specific cell surface antigens such as CD34, and the separation of erythroid progenitor cells based on binding to lectins such as Ulex europaeus agglutinin. After dissociation of erythroid progenitor cells, the lectin can be eluted from the cells by incubation with an excess of sugar, such as fucose, which competes for cell binding sites on the lectin. The invention also provides a method for treating a human patient having an erythroid disorder by administering a cell population enriched for human erythroid progenitor cells. Further provided is a diagnostic method based on the enriched cell population. |
