abstract | (57) [Summary]nPeptide vaccines and related methods for generating protective immune responses in warm-blooded animals are disclosed. Peptide vaccines are dividable by covalently linking conformationally constrained reverse turn pseudo-forms, T cell stimulating peptides and conformationally constrained reverse turn pseudo-forms and T cell stimulating peptides. Contains a linker molecule. A conformationally constrained reverse-turn pseudo-form is capable of pseudo-determining the three-dimensional structure of the antigenic determinant of the native pathogenic protein and binding to B cells. Upon binding to B cells, the peptide vaccine is taken up intracellularly and cleavage of the cleavable linker releases the T cell stimulating peptide from the conformationally constrained reverse turn pseudo-form. The T cell stimulating peptide is then associated with class I or II MHC molecules and presented on the surface of B cells, inducing T cell activation and producing a protective immune response. |
