abstract PURPOSE: To separate high purity leukocyte containing limited amount of erythrocyte, blood platelet, protein, etc., in high yield, by treating blood or body fluid with a leukocyte-capturing material or 9 filter to capture the leukocyte, and treating the captured leukocyte with a hemolyzing agent. n CONSTITUTION: Blood or body fluid such as cerebrospinal fluid, lymph, etc., or a cell dispersion obtained by removing most part of erythrocyte from the blood, etc., is treated with a leukocyte-capturing material such as nylon fiber, glass beads, etc., or a leukocyte-separating filter having a mesh size of about 5μ. The capturing material or the filter containing captured leukocyte is treated with a hemolyzing agent such as 0.25W0.35% NaCl solution to disintegrate the erythrocyte, and washed with a heparin-containing physiological saline solution. A dispersion of leukocyte containing less than several % of erythrocyte in number can be prepared by this process. n COPYRIGHT: (C)1979,JPO&Japio
