New pyrrole based compounds, compositions, and methods are provided that inhibit the activity of glycogen synthase kinase (GSK3) in vitro and treat GSK3 mediated disorders in vivo. The methods, compounds and compositions of the present invention can be used in the treatment of diabetes, Alzheimer's disease and other neurodegenerative diseases, obesity, atherosclerotic cardiovascular disease, essential hypertension, polycystic ovary syndrome, syndrome X, traumatic brain injury, bipolar disorder, immunodeficiency or cancer. It can be used alone or in combination with other pharmaceutical actives in the treatment of disorders mediated by GSK3 activity. |
