titleOfInvention | Methods and kits for primer based multiplex amplification of nucleic acids |
abstract | New methods for the diagnosis or differential diagnosis of disease agents and secondary disease agents are disclosed. The disclosed method uses a novel amplification strategy called TemPCR to allow sensitive and specific amplification of target sequences from any disease agent and / or secondary disease agent for which the nucleic acid sequence is known. The TemPCR method employs at least one set of target enrichment primers and at least one pair of covalent target amplification primers (highly present) specific for the disease agent or secondary disease agent being detected (low concentration). At least one pair of said target enrichment primers comprises a binding sequence for said target amplification primers. Thus, the use of the TemPCR method allows the multiplex amplification reaction to be performed without the need for experimental optimization of the multiplex amplification parameters. Also disclosed are nucleic acid isolation and detection of target sequences for use with the TemPCR method. |
