abstract | FIELD: biotechnology. n SUBSTANCE: fecal sample of a patient is kept at 96% absolute alcohol for 30-60 min in the ratio of alcohol and feces of 1:1, centrifuged for 15-20 minutes, then the fecal sample is treated with 1% solution of sodium deoxycholate in a ratio of 1:1 for 30-60 minutes, centrifuged, the supernatant is removed, then the primary inoculation of precipitation in the medium with addition of 1% lactose and 0.5-1.0 mM arabinose is carried out, incubated in anaerobic culture apparatus for 24-48 hours, the grown colonies are applied to the standard discs impregnated with the chromogenic substrate - ortho nitrophenyl-β-D-galactosidase. When appearance of bright yellow colour of the disc the toxigenic strains of Clostridium difficile are diagnosed. n EFFECT: use of the claimed method enables to diagnose toxigenic strains of Clostridium difficile with high sensitivity and specificity, simplicity and accessibility. n 3 ex |
