abstract | The invention comprises methods, primers and hybridisation probes for identifying organisms by means of comparative genetic analysis and is characterised in that coding and non-coding areas and/or functionally significant areas of highly conserved genes, pseudogenes or homologues are amplified using the PCR and then genotyped and analysed. The comparison of coding and non-coding areas of highly conserved genes, pseudogenes or homologues ensures that a single oligonucleotide pair bonds to DNA sequences that are highly conserved between different species, hereby allowing the amplification of a gene segment that is identical for all of the species. The oligonucleotides include one or more gene areas with the greatest possible sequence differences between different species. The determination of the gene sequence of this highly polymorphous area in a subsequent reaction step enables the gene sequence to be allocated to a specific species. In particularly preferred variants of embodiments of the invention, oligonucleotide pairs are found that make it possible to amplify the highly conserved tumour suppresser gene PTEN/MMAC1, its pseudogene and their homologues. |
