abstract |
Applicants have identified a novel amplification method that uses two specific primers to clone both the 5' and 3' cDNA ends in a single reaction. This new method also uses the first strand of cDNA obtained after reverse transcription of mRNA rather than double stranded cDNA, further increasing efficiency of amplification. According to the invention the first strand of cDNA is self-ligated using a DNA ligase. |
