abstract | The present invention relates to small organic molecules comprising at least one DNA-binding unit capable of sequence specific binding to the minor groove of a DNA molecule comprising the sequence 5'-GGG-3', said compound comprising covalently-linked first and second DNA-binding sub-units, each sub-unit comprising heterocyclic amino acid residues, the amino acid residues of the first sub-unit forming binding pairs with a portion of the amino acid residues of the second sub-unit, thereby providing a paired DNA-binding portion and an adjacent unpaired DNA-binding portion, wherein the paired portion and the unpaired portion together provide at least three consecutive amino acid residues preferentially binding to G, the paired portion providing no more than two consecutive residues preferentially binding to G. The invention also relates to the use of these compounds in targeting specific sequences, particularly telomeres, in vivo, in vitro and ex vivo. |
