abstract | The present invention relates to a hybrid pyrrolysyl tRNA (PylT), which is highly orthogonal and active. The invention furthermore provides a complex comprising a hybrid pyrrolysyl tRNA (PylT) and a pyrrolysyl-tRNA-synthetase (PylRS) (PylT/PylRS), as well as a pair of PylT/PylRS complexes, which are mutually orthogonal and active in the same system. The present invention allows for incorporation of at least one, such as two distinct, non-canonical amino acids (ncAA) into a protein. |
